I've heard many good things about LoL and have a few friends who play. Now that I have a lot of spare time over the summer, I'm thinking of giving it a try. Any tips for a newbie? What are some things I should definitely know? What are some things that I should definitely do/not do?

One important tip that I can give you, and most people will give you, is learn to "farm" minions. It's part of the tutorial too. You should try to deal the killing blow to minions so that you can get the gold from them.

It's probably best to practice in the co-op vs ai mode before you play actual PvP with other players.

Like Burt Povski pointed out well, learning to farm is crucial in LoL, as your game depends on items, and the most reliable alternative beside killing other players is to get constant gold from minions.
My other suggestion would be to learn getting by others, meaning that you shouldn't blame your support if they didn't do well. Instead, try to support him by saying he can do much, much better!
You shouldn't instalock, in my opinion. Have others know that you want to play a specific role, but don't choose your champion and lock the next second, because others will flame and blame you, and your game is screwed haha.
    Good luck! :)

Keep in mind as well that sometimes you'll be playing with players who AFK, so you'll be at a disadvantage of being 4v5. If that's the case, don't give up too easily. Sometimes, you can still with even if you are outnumbered.

    Here's some tips;

First have fun. Try not to take the game too seriously, don't let people get to you. Play co-op vs ai vs intermediate bots when you can, this is the best way to get used to the game and to face strong opponents without ruining a normal game with inexperience. last hitting is when you hit something just before its about to die so you get the money from it. to help with last hitting, press the S key, as it will stop your attack animation so you can time last hitting properly. this'll help you "CS", since you're new try to hit an average of at least 50cs by 10minutes. as you grow experienced in the game an average of around 70cs at 10minutes is what you'll want to aim for. Learn when to buy items. press P to open the shop, B to recall to the base. You want to effectively buy items, and by this I mean you return to base when you have enough gold for a part of an item. DO NOT HOLD ONTO A LOT OF GOLD FOR NO REASON. (ie: a item cost 3300 gold. don't wait until you have 3300 gold. the item is in parts, you can likely buy pieces at 1550, 875 and 730gold.)

Save your IP until you're level 20, do not buy ANYTHING. This includes champions unless you're 100% sure you want to play it. At level 20 you can buy tier 3 runes (The only runes worth buying in the game.) So you want to keep all of your IP so you have enough to fill out at least 2 rune pages towards the type of champions you like to play. by this time you should have found some guides (like on lolking or probuilds) to emulate and to help you fill it out.

2nd. lolking and probuilds. probuilds is obviously, where you can see what pro players build for runes/masteries on champs, good starting place. lolking, if you use the search bar you can find guides for champions (as well as look up other players.) try to use guides from people with 2000Lolkingscore and above as they are high in skill level and have good ideals on what they're talking about.

    good luck.

No problem Jesse, I hope it helps :D it's a lot to take in but there is no rush when it comes to league. Just take your time playing the game and use each match as a learning opportunity and you'll grow as a player in no time. :)

The best tip: Have fun. Some other ones I wish I knew when I first started: Try to improve, not to win every game. This will hopefully stop you from getting mad at people who suck or troll. Pick one or two champions for every role and practice them. At low levels and ELOs you can exploit so many mistakes in lane. If you're playing support, buy wards and use pings. Keep your ADC alive. No matter how inexperienced, they can still right click.

First of all, be prepared for some intense fights. You have to focus on the game because unlike dota, LOL is overpower. What I mean overpower? Fights between champions can be started as early as no creeps yet. Champions damage are so overpower that they have critical hits. Like master yi, if ignored, he can kill any champion for not more than 5 hits.

Don't give up too quickly, it might take some time to get where you want to be but just keep going.

All the tips in here so far are great! Absolutely, never forget this is a fun game, and you're playing it because you want to have fun! Whenever people get intense, or flame you (get mad at you for doing poorly) just remember you're learning and it's just a game.
The learning curve sorts itself out just from your time spent playing the game.

Haha that is one thing that cracked me up. People do get mad at it as if they're taking it too seriously which kills the fun in the game. I'd rather join people who are patient and don't take it way too seriously that they would get so angry and want to smash their PC. I agree with you, takes practise and learning.

Try out as many characters as you can, and enjoy yourself. Look up builds on probuilds.net and don't use mobafire, bad site with terrible ideas.
One character you can play as often as possible is someone you can main, and just play them non-stop, learn the ins and outs of them. It helps you learn the game, and learn the enemy characters as a whole.
League can best be described as chess mixed with basketball, you choose your players, but objectives stay the same. So learn what kind of trades you can have against the enemy team, if they get a kill, get a dragon, or a tower.

Try all the characters and experiment on them. I suggest you watch pro games so that you can you can learn how professionals play the game.

One of the most important things to do is learn from your mistakes and avoid falling into the same bad situations.

One of the best things about League is that no two games are exactly the same, and you need to be able to adapt to different situations quickly before you fall behind. The overall aim of the game is always the same (poet and I didn't know it), but your short term objectives aren't always going to be as exact.
